Again Im not hooked up to any thing yet only g540 and a battery just trying to get a voltage change within A set rpm. I do not want to move on until I know the g540/vfd 1-10v output works and  then I can move forward.

A couple things I would try, but I don't know if it matters.
1) set up an output and assign it to M3 clockwise. You probably need this to start the spindle. Are you calling M3 in Mach3 to start the spindle?
2) On the Spindle tab of ports and pins, uncheck the boxes in Special Functions. You have it trying to use feedback with out any feedback connected.
3) Make sure you have the correct pulley speed set up for the current pulley.

in"Ports and Pins" " Motor outputs"  Spindle  "Dir lowActive" has to be checkted and "SteplowActive" unchecked, you have it reversed.
Is working here tested with your profile.

Checked Pulleys verified 1:1  0-5000rpm and also 1-5000
Tried M3 S5000, 2500 and 1000 still get 6.5 volts
unchecked special boxes same outcome then  rechecked spindle averaging to show rpm range
Reversed dir low active and step low active still no change

What I did notice that might not mean anything is that as I kept the meter hooked up and went back to check it, I found that it is increasing voltage at a very slow rate fro 6.51 to 6.72 and climbing as it sits. could this just be residual or is there a frequency that could be raised I have 50hz in ss and in mach set the same. I believe the g540 is setup for 50hz.

In MotorTuning reduce the steps per to 50 and set the spindle acceleration  to max.

I did change the motor tuning settings several times and did try your 50 steps per and maxed out acc.
Maybe my g540 vfd I/O is toast because i get no change  6.9vdc when i should at least be getting at max rpm (5000) 9volts or just a change when spindle is off m3/m5

When you reversed dir low active and step low active ( see pict. ) using  your attached profile it should work.
When Mach is in Reset state there must be 0V with this settings.
