Assuming you require accurate X and Y axis machine movement replicating the pen movement from a tablet…
There is no current facility for it to be done within Mach but it could be possible if the output from a graphics tablet was compatible with, just for example, the output from a CNC Pendant and suitable drivers were present.

Certainly an interesting possibility for anyone adept at writing the driver software plugin.
