What is the best sure fire way to back up Mach3 settings?

I copy the entire Mach3 folder to a memory stick.

As Tom has said, make a backup of the whole Mach3 folder.

I even copy the desktop shortcuts as i use two profiles, whole M3 folder on a stick and laptop.

I have a copy of the mach installer on the stick too.
