I thought I would post here, in order to document the same problem I have had with a UC300 motion controller after an upgrade to windows 10
When I loaded mach3, it would say Plug in DLL defective. Reload"
I followed the advice on this forum and disabled the driver signature protocol as described. Still no luck.
I saved my xml file and uninstalled Mach3 and I uninstalled the driver for the UC300 as well
I reinstalled Mach3 and installed the automatic installer application for the uc300 from https://www.cnc4you.co.uk/UC300-USB-Motion-Controller
On that page they also have a download called "Software prerequirements" which is a copy of the dotnet framework for windows
I installed that and loaded Mach3 again and it worked

What worked for me was to go to Programs and Features (appwiz.cpl), choose Turn Windows Features on or off, then check the box for .NET Framework 3.5 (Includes .NET 2.0 and 3.0).

I was getting the same error "Plugin  DLL Defective. Reload." and "UC100 V2_151.dll - Defective Plugin Found..ignoreing.." on 64-bit Windows 10.
I used both driver options from the cncdrive website, and I had verified that the driver was installed properly, and that I had .NET installed (PowerShell reported 4.8.04084). The forums suggest that this DLL error is certainly related to .NET, so I'm guessing that while .NET 3.5 includes .NET 2.0 (required for the motion controller), higher versions do not. Making sure that .NET 3.5 was enabled included .NET 2.0, and now I'm in business!

wmgeorge I know, but it came up while I was searching for my own issue so I thought maybe it would still help others in my same boat!  ;D
