If the machine is fitted with home switches then all you need to do is jog the machine away from the limits and click "Ref All Home", machine should then go to the home position and stop.

Has the machine ever moved correctly?

What type of machine is it, Mill, Lathe, Router, 3D Printer

Are the axis direct drive?

Is it driving ball/lead screws, belts, rack and pinion?

Some pictures would help along with more info.

Stop procrastinating and fix you machine mechanics before you go any further.

In most cases if you have power to the Stepper drivers and the step and direction pin outs are correct then it is the enable line to the stepper drivers that is causing the issue.  When you enable the drives the motors will be very hard to near impossible to turn.

Some pictures of the setup showing the drives and UC100 connections will help diagnose the issue.

First are the motors energised, e.g. can you turn them by hand when powered up.

If you can :-

Do you have the enable on the drivers connected?

If you do remove them.  If not connect them and make sure the uc100 is controlling them.

Is this a new build/machine or has it been run before with out this problem.

If its new I would slow down the Z axis and give it moor acceleration time, it could be the head is missing steps on the rapid up moves.

Are the Z axis slides well lubricated?

The red leds will go green after you have homed the machine, the dropping is another issue, could be the steppers powering down or dodgy code in the M6 macro.
