Hello,
I have a problem, I connected the serial port to breakout board, but when I run Mach3, there's no signal sending out through the breakout board, nor the serial port. Any one knows the reason?

You need to use the parallel port rather than the serial port.
If you are using the parallel port then check you have the correct address set in Mach for it.
